Safety, tolerability, and efficacy of RIPK1 inhibitor, SAR443820, in amyotrophic lateral sclerosis (HIMALAYA): a multicentre, randomised, double-blind, placebo-controlled, phase 2 trial.

The Lancet. Neurology·October 1, 2026·PMID 42716044

Why it matters

RIPK1 inhibition was a mechanistically promising approach for ALS, but this well-powered trial found no functional benefit and a concerning hepatic safety signal. These results definitively close this therapeutic avenue and reinforce the importance of rigorous phase 2 testing before advancing neuroprotective candidates.
