Preclinical and first-in-human evaluation of VG-3927/SAR448851, an oral small-molecule TREM2 agonist for Alzheimer's disease.

Science translational medicine·October 14, 2026·PMID 42873892

Why it matters

Microglial dysfunction and TREM2 loss-of-function variants are major contributors to late-onset Alzheimer's disease. An oral TREM2 agonist that penetrates the CNS and engages the target could address a key unmet need, especially for patients with disease-associated TREM2 variants. The phase 1 data support further clinical development.
